You either choose open file if you have the base saved
Or 'new from clipboard' if you've just copied it. 

Brushes are something you have to experiment with, but definitely have the pixels size setting on smallest as you need to work on a small base

How do I remove the shadow on the pencil? Like when I draw there's like shadowing all around the px...

You can't, but you can use the Pixel pen in the menu(not the pen menu but the tool menu)
